This study reveals that conserved Z-DNA-forming sequences (Z-flipons) in human and mouse promoters enhance transcription reinitiation rates without affecting elongation, particularly in neurodevelopmental genes. Using deep learning (DeepZ) and experimental validation, the authors demonstrate that Z-flipons dynamically regulate gene expression by resetting transcription complexes or recruiting repressive chromatin machinery.
